“Pullulanase was extracellularly produced with an engineered Escherichia coli with a combined strategy. When auto-induction

instead of isopropyl beta-D-1-thiogalactopyranoside (IPTG) induction method was implemented, we observed increased extracellular activity (4.2 U ml(-1)) and cell biomass (7.95 g DCW l(-1)). Subsequent investigation of temperature effect on fermentation showed cultivation performed at 25 degrees C presented the highest extracellular titer and cell biomass. In order to reduce the extended production period, we developed a two-stage temperature control strategy. Its application not only reduced the production period from 72 to 36 h, but also further enhanced the yield of extracellular DAPT inhibitor pullulanase. Finally, with a view to releasing more intracellular pullulanase, we altered cell membrane PLK inhibitor permeability with various medium additives. As a result, extracellular titer was elevated to 68.23 U ml(-1), nearly 35-fold higher than that with IPTG induction method. The

combined strategy developed here may be useful for the production of other extracellular proteins by recombinant E. coli.”
“Biological properties of clove have been reported, but little is known about its effect on the immune system. This work was aimed to investigate the effect in vivo of a water-soluble part of hydroalcoholic extract of clove on pro-inflammatory cytokines (IL-1 and IL-6) production by macrophages of BALB/c mice. The action of the essential oil of clove on the production of these cytokines macrophages was also investigated in vitro. The chemical compositions of the extract and of the oil were also investigated. Treatment of mice with water extract of DNA Damage inhibitor clove was found to inhibit macrophages to produce both IL-1 and IL-6. The essential oil of clove also inhibited the

production of these cytokines in vitro. Eugenol was found to be the major component of the clove extract and essential oil, and probably is the causative agent of cytokine inhibition. Taken together, these data suggest an anti-inflammatory action of this spice.”
“The natural pregnane steroid 16-dehydropregnenolone (16-DHP) showed the growth inhibitory activity against human tumor cells; however, the mechanisms of actions of 16-DHP were not clarified. In this study, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay was used to investigate the growth inhibitory effect of 16-DHP. Cell cycle distribution was analyzed using flow cytometry. Hoechst 33258 staining and DNA agarose gel electrophoresis were used to detect apoptosis. The levels of proteins were probed by Western blotting, and caspase activities were analyzed using Caspase Activity Kit.
